The human telomerase reverse transcriptase (hTERT) promoter is a regulatory DNA sequence that controls the expression of the hTERT gene. In cancer therapy, it is exploited to drive tumor-selective replication of oncolytic viruses or expression of therapeutic genes due to its high activity in cancer cells and low/absent activity in most normal somatic cells. Activating mutations within this promoter are also frequently observed across a range of cancers.
