Overview

HUS1 checkpoint clamp component B (HUS1B) is a protein closely related to the canonical HUS1 protein, which is a component of the heterotrimeric 9-1-1 checkpoint clamp (composed of RAD9, RAD1, and HUS1). The 9-1-1 complex is structurally similar to proliferating cell nuclear antigen (PCNA) and plays a crucial role in the cellular response to DNA damage by participating in the activation of the DNA damage checkpoint and promoting cell cycle arrest and DNA repair[2][5]. HUS1B, however, is a paralog of HUS1, and while it can interact with RAD1, it does not form the canonical 9-1-1 clamp complex. Its precise function is unclear, and there is no evidence for its role as a validated therapeutic target, receptor, or enzyme. There are no known drug interactions, mechanisms of drug action, or established roles as a biomarker or safety concerns associated specifically with HUS1B[2][5].
