Overview

Hyaluronan scavenger receptors, primarily comprising Lymphatic Vessel Endothelial Hyaluronan Receptor 1 (LYVE-1) and Hyaluronan Receptor for Endocytosis (HARE, also known as Stabilin-2), are specialized glycoproteins that regulate the homeostasis of hyaluronan (HA) and other systemic glycosaminoglycans (Jackson, 2018; Zhou et al., 2003). LYVE-1 is predominantly expressed on lymphatic endothelial cells, where it acts as a docking receptor for HA-coated dendritic cells and macrophages, facilitating their entry into the lymphatic system for immune surveillance (Jackson, 2018). HARE/Stabilin-2, along with the related receptor Stabilin-1 (CLEVER-1), serves as a major clearance receptor for circulating HA and other ligands like heparin and oxidized LDL, primarily within the sinusoidal endothelium of the liver and lymph nodes (Harris & Weigel, 2008; Schledzewski et al., 2011). These receptors are significant therapeutic targets in oncology and inflammatory diseases, as they facilitate tumor-induced lymphangiogenesis and the metastatic spread of cancer cells to lymph nodes (PNAS, 2011; MDPI, 2021). Therapeutic strategies include monoclonal antibodies, such as bexmarilimab (targeting Stabilin-1), to modulate the immune microenvironment, and HA-conjugated delivery systems that exploit receptor-mediated endocytosis for targeted drug delivery (Novus Bio, 2024; ResearchGate, 2025).
