Overview

Hyaluronan synthase (HAS) refers to a family of membrane-bound enzymes responsible for the biosynthesis of hyaluronan (HA), a major component of the extracellular matrix. In mammals, there are three main isoforms: HAS1, HAS2, and HAS3. These enzymes catalyze the polymerization of HA by sequentially adding UDP-α-N-acetyl-D-glucosamine (UDP-GlcNAc) and UDP-α-D-glucuronic acid (UDP-GlcA) to form high molecular weight HA chains. HAS enzymes play critical roles in tissue homeostasis, wound healing, and inflammation, making them potential therapeutic targets for diseases such as cancer and fibrosis.
