Overview

Hyaluronan synthase 2 antisense RNA 1 (HAS2-AS1) is a long non-coding RNA transcribed from the opposite strand of the *HAS2* gene locus on chromosome 8q24.13[1][3]. As a cis-encoded natural antisense RNA, it forms heteroduplexes with *HAS2* mRNA and regulates *HAS2* gene expression both in the nucleus—by altering chromatin structure—and in the cytoplasm—by modulating mRNA stability[1][3]. HAS2-AS1 exists in at least two splice variants and coordinates with various signaling pathways, such as those induced by interleukin-1β, TGF-β1, LH/hCG, and transcription factors like Sp1, Smad2/3, C/EBPβ, and USF1[1][2][4][6]. Functional studies show HAS2-AS1 enhances cell proliferation and migration and modulates inflammatory cytokine expression, supporting roles in cancer progression, fibrosis, and pulmonary arterial hypertension[4][5][6]. It regulates disease-related cellular phenotypes but is not currently known as a direct therapeutic target for approved drugs or a classical "receptor, enzyme, or transporter"[3][5]. However, it may serve as a biomarker for aggressiveness in breast cancer and glioma, and as a potential research target for therapeutic intervention in fibrosis and cancer[5][6]. No interacting drugs or safety concerns are currently established.
