Overview

Hyaluronidase 4 (HYAL4) is an enzyme encoded on human chromosome 7q31.3 that acts as a chondroitin sulfate-specific endo-beta-N-acetylgalactosaminidase, cleaving chondroitin sulfate chains predominantly of the CS-D motif and playing a role in extracellular matrix metabolism. Unlike other hyaluronidases, its substrate range is more restricted and sequence features set it apart from other family members. HYAL4 is expressed in several tissues, with enrichment in placenta, skeletal muscle, and testes, and is implicated in normal developmental processes and disease states, including a variety of cancers and joint or neural tissue injury. Its activity regulates cell surface proteoglycans such as serglycin, aggrecan, CD44, and sulfatase 2. Recent research suggests roles in cancer biology, stem cell regulation, tissue development, and possible therapeutic potential as a disease biomarker or drug target, although selective modulators and clinical applications remain underdeveloped.
