Hydroxyacid oxidase 2 (HAO2)

Target
HAO2
Molecular classification
Enzyme, Oxidase, Metabolic protein, Peroxisomal protein
01

Overview

Hydroxyacid oxidase 2 is a peroxisomal enzyme that catalyzes the oxidation of medium and long-chain 2-hydroxyacids such as 2-hydroxyhexadecanoate and 2-hydroxyoctanoate into their corresponding 2-oxoacids, using O₂ as the physiological electron acceptor, and thereby produces H₂O₂. It plays a key role in the alpha-oxidation pathway of fatty acids. Loss or downregulation of HAO2 has been implicated in tumorigenicity of hepatocellular carcinoma, suggesting a tumor suppressive function. Alternative splicing results in multiple transcript variants and the enzyme is not active on glycolate or small chain substrates.

Cancer (notably inhibits tumorigenicity in hepatocellular carcinoma)Inborn errors of metabolism (e.g., Histidinemia, Dimethylglycine Dehydrogenase Deficiency)Other (possible roles in metabolic syndromes)
05

Safety considerations

As HAO2 is not a direct drug target, specific safety concerns are presently undocumented.General challenges could relate to modulation of peroxisomal oxidases or hydrogen peroxide generation, potentially impacting oxidative stress and cell viability.
