Hydroxymethylglutaryl-CoA (HMG-CoA) is an intermediate in the mevalonate pathway, a crucial metabolic pathway responsible for the synthesis of cholesterol, isoprenoids, and other essential biomolecules. It is formed from acetyl-CoA and acetoacetyl-CoA through the action of HMG-CoA synthase. HMG-CoA itself is not a direct therapeutic target, but it is a precursor to mevalonic acid, the synthesis of which is regulated by HMG-CoA reductase, a well-established drug target for cholesterol lowering.
