Overview

Hydroxysteroid 17-beta dehydrogenase 12 (HSD17B12) is a multifunctional enzyme in the short-chain dehydrogenase/reductase superfamily that catalyzes two critical metabolic processes: the conversion of estrone to estradiol (an essential step in estrogen biosynthesis) and the elongation of long- and very long–chain fatty acids, particularly involved in arachidonic acid production[1][2][3]. The enzyme is crucial for embryonic development, reproductive physiology, and neuronal development. In cancer biology, HSD17B12 is increasingly recognized as a marker of aggressive disease, particularly in breast and ovarian cancer, where its overexpression correlates with poor clinical outcomes and its silencing impairs tumor cell proliferation. Genome-wide association studies implicate HSD17B12 in multiple tumor types and reproductive disorders, given its roles in steroidogenesis and lipid metabolism[1][2].
