Hydroxysteroid 17-beta dehydrogenase 13 (HSD17B13) is a liver-enriched enzyme belonging to the short-chain dehydrogenase/reductase (SDR) superfamily. It plays a crucial role in hepatic lipid metabolism by catalyzing the oxidation of various lipid substrates, including steroids, eicosanoids, and retinoids. HSD17B13 is primarily localized to lipid droplets within hepatocytes and its expression is significantly upregulated in nonalcoholic fatty liver disease (NAFLD). Loss-of-function variants of HSD17B13 have been shown to protect against hepatic steatosis, fibrosis progression, and chronic liver disease, making it a promising therapeutic target for NAFLD and related conditions. Therapeutic strategies involve inhibitors or RNAi-based therapies aimed at reducing hepatic HSD17B13 expression.
