Overview

Hydroxysteroid 17-beta dehydrogenase 14 (HSD17B14) is a mammalian enzyme belonging to the short-chain dehydrogenase/reductase (SDR) family, primarily known for its role as L-fucose dehydrogenase—the initial enzyme of the L-fucose degradation pathway[1][2][3][5]. HSD17B14 catalyzes the NAD(+)-dependent oxidation of L-fucose to L-fucono-1,5-lactone, which then spontaneously rearranges to L-fucone-1,4-lactone[1][5]. Its substrate specificity includes L-fucose as the preferred substrate, with lower catalytic efficiency for D-arabinose and L-galactose[5]. The enzyme may play additional metabolic roles involving steroid, fatty acid, and prostaglandin metabolism, but its physiological functions—especially in the context of L-fucose degradation—remain incompletely understood[1][3]. No direct therapeutic applications, drug interactions, or disease associations are currently established for HSD17B14.
