Hypermethylated in cancer 1 (HIC1), also known as HIC ZBTB transcriptional repressor 1, is a transcriptional repressor protein belonging to the ZBTB family, characterized by a BTB/POZ domain and C2H2-type zinc finger motifs[4][1][2][3]. HIC1 functions mainly as a tumor suppressor and growth regulator, frequently inactivated in cancer through promoter hypermethylation or chromosomal deletion[4][7]. Its transcriptional repression activity inhibits gene expression by binding to DNA and recruiting corepressors and histone-modifying enzymes via its BTB domain[1][2]. HIC1 is implicated in maintaining genomic integrity and regulating responses to DNA damage. Loss or silencing of HIC1 is recognized in the pathogenesis of multiple cancers, including breast, colorectal, and brain tumors[4]. It is not considered a direct therapeutic target (not a receptor, enzyme, or transporter), but its inactivation status is a relevant cancer biomarker.
