Overview

Hyperpolarization-activated cyclic nucleotide-gated (HCN) channels are nonselective voltage-gated cation channels primarily found in heart and brain cells. They are encoded by four genes (HCN1-4) and play a crucial role in controlling rhythmic electrical activity, such as cardiac pacemaking and neuronal excitability. HCN channels activate upon hyperpolarization and are further modulated by intracellular cyclic nucleotides (cAMP, cGMP, cCMP). Dysfunction of HCN channels is implicated in cardiac arrhythmias and neurological disorders, making them a therapeutic target for conditions like bradycardia, epilepsy, neuropathic pain, and depression.
