Overview

Candida albicans Hyphally regulated cell wall protein 1 (Hyr1) is a glycosylphosphatidylinositol (GPI)-anchored surface protein expressed specifically during the hyphal (filamentous) growth phase of the fungus. It serves as a critical virulence factor by mediating resistance to host innate immunity, specifically protecting the fungus from neutrophil-mediated phagocytosis and killing. Hyr1 has also been identified as a cross-kingdom receptor that interacts with outer membrane proteins of Gram-negative bacteria like Acinetobacter baumannii, facilitating the formation of mixed-species biofilms and enhancing bacterial virulence. Due to its surface exposure and essential role in pathogen survival and immune evasion, Hyr1 is a primary candidate for antifungal vaccines and monoclonal antibody therapies. Experimental vaccines utilizing the recombinant N-terminal domain (rHyr1p-N) have demonstrated protective efficacy in animal models against disseminated candidiasis and lethal bacterial infections. Therapeutic strategies targeting Hyr1 focus on neutralizing its protective function, thereby sensitizing the pathogen to host immune responses and disrupting the structural integrity of biofilms in clinical settings.
