Overview

The hypothalamic–pituitary axis is a central regulatory system that integrates the functions of the hypothalamus and the pituitary gland to maintain endocrine homeostasis. The hypothalamus acts as the command center, secreting neurohormones that travel through the hypophyseal portal system to the anterior pituitary, where they stimulate or inhibit the release of tropic hormones (StatPearls, 2023). These pituitary hormones, such as ACTH, TSH, and LH/FSH, then regulate the activity of peripheral target glands including the adrenal glands, thyroid, and gonads (NIH, 2022). This axis is critical for managing the body's response to stress, growth, metabolism, and reproductive cycles. Dysregulation of the axis can lead to a wide range of disorders, such as Cushing's syndrome, hypothyroidism, and growth hormone deficiency (Wikipedia, 2024). Pharmacological management typically involves replacing deficient hormones or using analogs to modulate specific feedback loops within the axis (PubMed, 2021). Because it represents a complex physiological pathway involving multiple organs and signaling molecules, it is classified as a system rather than a single molecular drug target.
