Hypothalamic-pituitary-gonadal axis is a critical endocrine system comprising the hypothalamus, anterior pituitary gland, and gonads (ovaries or testes), which together regulate reproduction, development, gonadal steroid production, and aging. The axis operates via feedback loops: hypothalamic GnRH secretion prompts pituitary release of LH and FSH, stimulating the gonads to produce sex steroids (estrogen, testosterone, progesterone), which in turn modulate upstream axis activity. The system promotes sexual maturation, gamete production, menstrual/ovarian cycling in females, and spermatogenesis and androgen synthesis in males. Dysfunction or deregulation of the HPG axis underlies conditions such as precocious puberty, delayed puberty, infertility, hypogonadism, and has roles in reproductive cancers.
