The hypothalamic thermoregulatory center is a critical neural circuit located primarily in the preoptic area (POA) of the anterior hypothalamus. It acts as the body's thermostat, integrating sensory input about internal and external temperatures to maintain a stable core body temperature. The center orchestrates physiological responses such as vasodilation/vasoconstriction, sweating, shivering, and behavioral changes to maintain thermal homeostasis. Key neuronal types include heat-sensitive GABAergic neurons in the POA and cold-sensitive glutamatergic/GABAergic neurons in the dorsomedial hypothalamus (DMH). TRP channels, like TRPM2, are implicated as potential molecular sensors. The center also regulates thyroid hormone release via TRH, affecting metabolic rate and heat production. Its proper function is essential for survival, and disruption can have fatal consequences.
