Overview

The hypothalamus-pituitary-ovarian axis (HPO axis) is a tightly regulated physiological system that coordinates menstrual cycle and female reproductive function via hormone signaling between the hypothalamus, pituitary gland, and ovaries. The hypothalamus secretes gonadotropin-releasing hormone (GnRH), stimulating the pituitary to release luteinizing hormone (LH) and follicle-stimulating hormone (FSH), which then act on the ovaries to regulate production of estrogen and progesterone. This communication occurs in feedback loops, controlling follicle development, ovulation, and endometrial changes. Dysfunction or disease affecting any portion of the axis can lead to ovulatory disorders, infertility, or other health outcomes. The axis itself is the focus of various therapies aimed at restoring or modulating reproductive hormone states.
