Hypoxia-inducible Factor 1 Beta (HIF-1β)

Target
HIF-1β
Molecular classification
Transcription factor, Basic helix-loop helix-PAS family
01

Overview

Hypoxia-inducible factor 1 beta (HIF-1β), also known as aryl hydrocarbon receptor nuclear translocator (ARNT), is a constitutively expressed subunit of the HIF-1 transcription factor complex. HIF-1 is a heterodimeric protein composed of an oxygen-regulated alpha subunit (HIF-1α) and the constitutive beta subunit (HIF-1β/ARNT). This complex plays a central role in cellular adaptation to hypoxic conditions by regulating the expression of genes involved in oxygen homeostasis, metabolism, angiogenesis, and cell survival. ARNT can dimerize with other members of the bHLH-PAS family beyond just HIF-alpha isoforms.

Mechanism of action

Not applicable as HIF-1β/ARNT is a constitutively expressed subunit. Therapeutic strategies focus on targeting the oxygen-sensitive alpha subunits of HIF-1 or downstream targets.

Safety considerations

Inhibition of HIF-1β could have broad effects on cellular processes due to its role in multiple pathways.Potential for off-target effects due to the bHLH-PAS domain being present in other proteins.
