Overview

Hypoxia-inducible factor 2-alpha (HIF2α) mRNA is the messenger RNA transcript encoded by the EPAS1 gene, serving as the essential template for the synthesis of the HIF2α protein. This protein acts as a master transcription factor that orchestrates the cellular response to low oxygen levels by activating genes involved in angiogenesis, erythropoiesis, and cell survival (UniProt P42330). In pathological contexts, particularly clear cell renal cell carcinoma (ccRCC), the loss of the von Hippel-Lindau (VHL) tumor suppressor leads to the constitutive stabilization of HIF2α, driving aggressive tumor growth and vascularization (NCBI Gene ID: 2034). While small molecules like belzutifan target the protein product, therapeutic strategies targeting the HIF2α mRNA utilize RNA interference (siRNA) or antisense oligonucleotides (ASOs) to degrade the transcript and prevent protein production entirely. Clinical candidates such as ARO-HIF2 and IONIS-AR-2.5-Rx have been developed to exploit this mechanism, offering a targeted approach for VHL-deficient malignancies and other hypoxia-driven disorders (PubMed: 31511301).
