H-NOX (Heme-Nitric Oxide/OXygen binding) proteins are a family of heme-containing sensor proteins that bind nitric oxide (NO), and in some cases, oxygen (O₂). They function as sensors for these small gaseous molecules, triggering conformational changes that initiate downstream signaling pathways. Engineered or naturally occurring O₂-binding variants of H-NOX could theoretically be used for targeted oxygen delivery under hypoxic conditions due to their ability to bind/release O₂ depending on local environment cues. Their tunable ligand affinity makes them attractive candidates for hypoxia-targeted therapies.
