I-Ag7 is a murine MHC class II molecule strongly associated with susceptibility to type 1 diabetes in NOD mice. It presents peptides derived from pancreatic beta cells to CD4+ T cells, initiating autoreactive immune responses. Its unique structural features, particularly polymorphisms in the beta chain (His56/Ser57), alter its peptide binding specificity, favoring peptides with acidic residues at P9 and enhancing its interaction with diabetogenic T cell receptors.
