I-kappa B kinase beta (IKK-beta) is a serine/threonine protein kinase and a key component of the IKK complex, which also includes IKK-alpha and NEMO. The IKK complex plays a central role in the canonical NF-kappa B signaling pathway. IKK-beta phosphorylates IκB proteins, leading to their ubiquitination and degradation, which in turn allows NF-kappa B dimers to translocate to the nucleus and activate gene transcription. This pathway is critical for immune responses, inflammation, and cell survival. IKK-beta is activated by pro-inflammatory stimuli such as TNF-alpha, IL-1, and LPS. Due to its role in inflammation and cancer, IKK-beta is a therapeutic target for inflammatory diseases, autoimmune disorders, and cancer.
Inhibition of IKK-beta leads to reduced phosphorylation of IκB proteins, preventing NF-kappa B activation and subsequent transcription of inflammatory and pro-survival genes.
