Overview

ID2 antisense RNA 1 (ID2-AS1) is a head-to-head antisense long non-coding RNA that is transcribed in the antisense direction relative to the ID2 gene. ID2-AS1 acts as a cis-acting regulator, promoting expression of the adjacent ID2 gene by interfering with the recruitment of histone deacetylase 8 (HDAC8) to the ID2 promoter. Its upregulation increases local chromatin accessibility, boosting ID2 expression—a factor associated with suppression of metastasis and inhibition of the epithelial–mesenchymal transition in hepatocellular carcinoma cells. Research suggests that ID2-AS1 functions as a tumor suppressor in specific cancer contexts by modulating gene expression via chromatin modification and transcriptional activation of the neighboring tumor suppressor gene ID2. If structured information is needed for bioinformatics or therapeutic exploration, ID2-AS1 is best classified as a regulatory non-coding RNA with supportive data for a role in cancer biology, but as of now, it is neither a druggable target nor a direct molecular target of approved therapies.
