Overview

The idiotype antigen on a clonal B cell receptor is the unique set of antigenic determinants located within the variable region of a B cell’s immunoglobulin receptor (BCR), which confers specificity for a particular antigen[1][2][3][4]. This unique region can be recognized as an antigen by other immune cells or by laboratory-generated antibodies (anti-idiotype), and forms the basis for idiotype-targeted immunotherapies in B cell malignancies. Because every B cell clone produces a distinct idiotype, the idiotype acts both as a fingerprint for malignant clonal populations in lymphoproliferative disorders and a target for highly specific therapeutic intervention. Idiotype–anti-idiotype interactions also play important roles in immune regulation and maintaining adaptive immune memory[4].
