Overview

The idiotype immunoglobulin refers to an immunoglobulin's distinctive set of antigenic determinants (idiotopes) found in the variable regions of its heavy and light chains. These arise due to gene rearrangement and somatic mutation during B cell development, endowing each antibody with a unique surface for antigen binding. Idiotypes are crucial in immune recognition and are exploited as therapeutic targets, particularly in B cell lymphoma, where each malignant clone expresses a unique idiotype that can serve as a tumor-specific antigen. They can also serve as surrogate antigens in vaccine approaches, leveraging anti-idiotype responses to induce immunity. However, their use poses challenges due to variability and the potential for immune-mediated adverse effects.
