Overview

IER3 antisense RNA 1 (IER3-AS1) is a long non-coding RNA transcribed from the strand opposite to the IER3 gene, sharing a 325 bp overlapping region with IER3. It interacts with its sense counterpart via RNA–RNA duplex formation, regulating the stability and expression of both transcripts in a context-dependent manner. This lncRNA is induced early in response to growth factors (such as FGF-2), localizes to both the nucleus and cytoplasm (with dominant nuclear localization), and its co-expression with IER3 has been shown to promote cell proliferation and a poor prognosis in several cancers. However, its function and impact are highly compartment- and cancer-type specific, as in neuroblastoma it does not exhibit oncogenic activity nor strong expression correlation with IER3. There are no direct drugs or therapies targeting IER3-AS1 as of now, but its expression and regulatory relationship with IER3 have implications for tumor biology and potential future therapeutic targeting of lncRNA networks.
