Overview

The IFN-gamma-secreting Th1 effector T cell polarization pathway describes the molecular and cellular processes by which naïve CD4+ T cells differentiate into Th1 effector cells, characterized by their ability to produce interferon-gamma (IFN-γ). This pathway is central to cell-mediated immunity against intracellular pathogens and plays a role in autoimmune and inflammatory diseases. Key molecules include IL-12, IL-27, IFN-γ, T-bet, and STAT1. The pathway involves initiation by pro-Th1 cytokines, differentiation through positive feedback loops involving T-bet and IFN-γ, and effector function primarily through IFN-γ secretion. Regulation occurs via factors like p38 MAP kinase and differential receptor expression. Th1 responses are critical for defense against intracellular pathogens but can also contribute to immunopathology.
