Overview

The IgE-allergen epitope repertoire represents the collective set of specific molecular regions on allergen proteins that are recognized and bound by Immunoglobulin E (IgE) antibodies. This repertoire is fundamentally heterogeneous, as it encompasses a wide variety of binding sites across different protein families (e.g., Bet v 1 from birch pollen or Ara h 1 from peanuts) and varies significantly between individuals based on their unique immune sensitization profiles (Sánchez-Trincado et al., 2017). The interaction between IgE and these epitopes is the primary driver of type I hypersensitivity reactions, as it facilitates the cross-linking of high-affinity IgE receptors (FcεRI) on effector cells like mast cells and basophils, leading to the explosive release of inflammatory mediators (Valenta et al., 2010). Therapeutic interventions often target this repertoire through allergen-specific immunotherapy (AIT) to induce desensitization or through the use of engineered monoclonal antibodies, such as the REGN5713-5714-5715 cocktail, which act as "blocking" agents to prevent IgE-allergen complex formation (Regeneron Pharmaceuticals). Because the repertoire is so diverse and often poorly defined for specific patients, characterizing the dominant epitopes is essential for the development of effective component-resolved diagnostics and targeted biologics in allergy management.
