Overview

IGF-like family receptor 1 (IGFLR1) is a structurally novel cell-surface receptor resembling members of the TNFR superfamily, encoded on chromosome 19. Its ligand is IGF-like family member 1 (IGFL1). IGFLR1 is expressed broadly, but especially in lymph nodes, spleen, kidney, and on T cells. Functionally, IGFLR1 binds IGFL1 with high affinity and participates in regulation of T cell responses, with elevated expression promoting inflammatory infiltration and activation. Recent evidence links IGFLR1 to pathogenesis and prognosis in several cancers, such as pancreatic ductal adenocarcinoma and clear cell renal cell carcinoma. High IGFLR1 expression is associated with poor survival in some tumor types, and its presence marks aggressive T cell phenotypes. As a novel therapeutic target, IGFLR1 currently lacks direct pharmacological modulators, but research is ongoing into its role in immune regulation and cancer biology.
