Overview

IGF1R antisense imprinted non-protein coding RNA (IRAIN) is a long non-coding RNA transcribed antisense to the IGF1R gene from an intronic promoter within the IGF1R locus, and is exclusively expressed from the paternal allele due to genomic imprinting[2][3]. Unlike the protein-coding IGF1R transcript, IRAIN does not code for a protein but plays a regulatory role in gene expression and chromatin architecture, including facilitating enhancer/promoter looping at the IGF1R locus[2][3]. IRAIN has tumor-suppressive properties and is found to be downregulated in leukemia cell lines and in patients with high-risk acute myeloid leukemia, where its lower expression is associated with more aggressive disease[2][4]. In multiple myeloma, IRAIN acts as a tumor suppressor in part by being regulated through miR-125b interactions[4]. Increasing IRAIN expression can downregulate IGF1R in cancer cells, further supporting its function as a suppressor of IGF1R-mediated oncogenic pathways[5]. There are currently no known drugs that directly target IRAIN, and its safety concerns as a therapeutic target have not been characterized.
