Overview

IGFL2 antisense RNA 1 (IGFL2-AS1) is a long non-coding RNA (lncRNA) located on chromosome 19. It is transcribed antisense to the IGFL2 gene, does not encode a protein, and has multiple splice variants[4][5]. IGFL2-AS1 is highly upregulated in several cancers, notably colorectal cancer, where it promotes tumor cell proliferation, migration, invasion, and epithelial–mesenchymal transition by modulating multiple signaling pathways, such as the Wnt/β-catenin pathway, HIF-1α/CA9 axis, and KRAS signaling[1][2][3][6]. Functionally, IGFL2-AS1 acts as a competing endogenous RNA (ceRNA), sequestering specific microRNAs and thereby influencing downstream target genes critical for cancer progression. High expression of IGFL2-AS1 correlates with worse clinical outcomes and advanced tumor stage, and is being explored as a prognostic biomarker in oncology[2][3]. No drugs directly targeting IGFL2-AS1 are currently in clinical use.

Mechanism of action

Not applicable for drugs; IGFL2-AS1 functions as a molecular sponge for microRNAs (e.g., miR-1224-5p, miR-802), modulating genes such as SATB1 and IGFL1, upregulating HIF-1α and CA9 by suppressing HIF-1α degradation, thereby activating signaling pathways involved in cancer progression[1][2][3][6]
