Target intelligence / Profile preview

IKAROS family zinc finger 1

Molecular classification
Transcription factor, Zinc finger protein
01

Overview

IKZF1, also known as Ikaros, is a zinc finger transcription factor crucial for lymphoid cell development and hematopoiesis. It regulates gene expression through chromatin remodeling and interacts with co-repressors and co-activators. IKZF1 is frequently deleted or mutated in B-cell precursor acute lymphoblastic leukemia (B-ALL), leading to poor prognosis. While direct targeting of IKZF1 is not currently possible, downstream pathways affected by its loss are being explored for therapeutic intervention.

Other names
Ikaros
02

Mechanism of action

Indirectly targetable via downstream pathways like JAK/STAT signaling or PI3K pathway. Casein kinase 2 inhibitors can restore some IKZF1 function.

03

Biological functions

Lymphoid differentiationRegulation of hematopoiesisTumor suppressionRegulation of gene expressionChromatin remodeling
04

Disease associations

Acute lymphoblastic leukemia (ALL)Immunodeficiency syndromes
05

Safety considerations

Loss of IKZF1 function leads to treatment resistance in ALL.Therapeutic targeting of downstream pathways may have off-target effects.
06

Biomarkers

IKZF1 deletions/mutations in B-ALL (prognostic biomarker)SNPs near IKZF1 (susceptibility to childhood ALL and type 1 diabetes)
