Overview

The Ikaros family zinc finger (IKZF) proteins, including IKZF1 (Ikaros), IKZF2 (Helios), IKZF3 (Aiolos), and IKZF4 (Eos), are a group of transcription factors essential for the development and regulation of the immune system. These proteins play critical roles in lymphocyte differentiation and the maintenance of immune homeostasis, particularly in the suppression of effector T-cell functions by regulatory T-cells (Tregs). In the context of oncology, IKZF proteins are often exploited by tumors to maintain an immunosuppressive microenvironment, where high levels of Helios and Eos in Tregs prevent robust anti-tumor immune responses. BMS-986482 is a novel small-molecule Cereblon E3 Ligase Modulator (CELMoD) designed to act as a pan-degrader of the IKZF1-4 family. By inducing the selective degradation of these transcription factors, the drug aims to reprogram the immune environment, reversing Treg-mediated suppression and enhancing the activity of effector T-cells against solid tumors.
