Immune system – non-specific immunological activation refers to a therapeutic strategy aimed at broadly enhancing the body's immune response rather than targeting a specific antigen or molecular pathway (NIH, 2022). This process typically involves the activation of the innate immune system, which provides a robust environment for the adaptive immune system to function more effectively against pathogens or malignant cells (NCI, 2023). Common agents used for this purpose include bacterial products like Bacillus Calmette-Guérin (BCG), which is used for bladder cancer, and cytokines such as Interferon-alpha and Interleukin-2 (PubMed, 2021). These agents work by stimulating pattern recognition receptors (PRRs) and inducing the production of various pro-inflammatory cytokines, thereby enhancing the activity of natural killer (NK) cells and macrophages (StatPearls, 2023). While effective in certain contexts, this non-specific approach can lead to significant safety concerns, including systemic inflammatory response syndrome and the potential for autoimmune reactions (Wikipedia, 2024). Because it describes a physiological process or therapeutic class rather than a specific protein or receptor, it is not considered a molecular target in the traditional sense.
The mechanism involves the broad stimulation of the innate immune system through the activation of pattern recognition receptors (PRRs), such as Toll-like receptors, and the subsequent induction of a cytokine cascade (e.g., IL-2, IFN-gamma) that enhances the cytotoxic activity of natural killer cells and macrophages (StatPearls, 2023; NIH, 2022).
