The IgE-mediated allergy pathway is a hypersensitivity reaction in which the immune system overreacts to harmless substances (allergens) by producing allergen-specific IgE antibodies. These IgE antibodies bind to high-affinity FcεRI receptors on mast cells and basophils, leading to rapid degranulation and release of inflammatory mediators upon re-exposure to the allergen, causing acute allergic symptoms. Strategies targeting this pathway include anti-IgE monoclonal antibodies that block IgE binding or therapies modifying glycosylation patterns on circulating IgEs.
