The immune system serves as the functional target for pneumococcal capsular polysaccharide–protein conjugates, which are designed to elicit a robust and lasting immune response against Streptococcus pneumoniae (CDC, 2023). These conjugates consist of bacterial capsular polysaccharides covalently linked to a carrier protein, such as CRM197 or diphtheria toxoid, to overcome the limitations of T-cell independent antigens which typically fail to induce memory in infants (Pollard et al., 2009). By converting the polysaccharide into a T-cell dependent antigen, the vaccine allows the complex to be processed by B cells and presented to T-helper cells via the Major Histocompatibility Complex (MHC) class II pathway (Pichichero, 2013). This interaction triggers B-cell class switching, affinity maturation, and the development of long-lived memory B cells and plasma cells (StatPearls, 2023). Clinically, these vaccines (e.g., Prevnar 13, Prevnar 20) are essential for preventing invasive pneumococcal diseases, including pneumonia, meningitis, and bacteremia, by generating high-affinity antibodies that facilitate the opsonophagocytosis of the pathogen (FDA, 2021).
