Mechanism of action

AS03 rapidly activates multiple innate immune pathways including interferon signaling, JAK-STAT pathway, Fcγ receptor (FcγR)-mediated phagocytosis, and antigen processing/presentation via MHC class I and II molecules. At the injection site, AS03 promotes local inflammation that recruits antigen-presenting cells (APCs) such as dendritic cells. This enhances antigen uptake and migration to draining lymph nodes where adaptive immunity is initiated. The presence of α-tocopherol in the formulation modulates the magnitude of the innate response by increasing cytokine production and improving antigen presentation efficiency. Downstream effects include robust memory B cell formation, increased CD4+ T cell activation, and higher total IgG titers with greater antibody avidity.
