Overview

The depot effect refers to the pharmacological phenomenon in which injected antigens or drugs are retained at a localized site, leading to prolonged exposure and sustained activation of the immune system. In vaccines, adjuvants such as aluminum salts, specialized polymers, or emulsions form a depot, slowly releasing antigen and recruiting immune cells to the injection site. This mechanism enhances antigen uptake by antigen-presenting cells (dendritic cells, macrophages), stimulates cytokine and chemokine production, and generates stronger and longer-lasting immune responses, both humoral (antibody) and cellular (T cell). The depot effect is a key principle in vaccine formulation but is not a discrete molecular target; rather, it is a property of certain delivery systems and adjuvant formulations that improve immunogenicity against cancer, infectious disease, and other indications. Safety challenges include potential for local inflammation, granuloma formation, or excessive immune activation.
