Immune system activation via release of tumor-associated antigens

Molecular classification
Immune process, Antigen presentation, T cell activation
01

Overview

Activation of the immune system via release of tumor-associated antigens is a foundational concept underpinning several modern cancer immunotherapies. It leverages natural processes whereby dying cancer cells provide a source of recognizable targets for both innate and adaptive arms of host defense. While challenges remain regarding specificity, potency, and overcoming tolerance/immunosuppression, this target remains central in ongoing efforts toward effective therapeutic vaccination and combination regimens across oncology indications.

Mechanism of action

Tumor-associated antigens are released from dying tumor cells, taken up by antigen-presenting cells (APCs), processed and presented on MHC molecules. This leads to the activation of T lymphocytes, particularly CD8+ cytotoxic T cells, which recognize and kill tumor cells expressing those antigens.
