Overview

Desensitization via gradual exposure to peanut proteins is a form of allergen-specific immunotherapy in which individuals with peanut allergy are given increasing oral doses of peanut protein under medical supervision to reduce immune system reactivity to peanuts. This approach downregulates the activity of certain T cell subsets (particularly TH2A cells), decreases the output of allergic cytokines (such as IL-4, IL-5, IL-9, and IL-13), and reduces activation of effector cells like basophils and mast cells. The primary goal is to safely increase the threshold for allergic reactivity and prevent severe allergic reactions from accidental exposures, although true immunological tolerance is rarely achieved and the effect usually persists only with continual exposure maintenance[1][2][3].
