Overview

Immune system effector cells are activated immune cells, primarily differentiated T cells (such as cytotoxic CD8+ T cells and helper CD4+ subsets like Th1, Th2, Th17), B cells (plasma cells), macrophages, and natural killer cells, that execute direct immune responses against pathogens, infected cells, or tumors. They arise from naive lymphocytes upon antigen encounter via T cell receptors (TCR) or B cell receptors (BCR), leading to proliferation, differentiation, and functional specialization; for example, cytotoxic T lymphocytes release perforin and granzymes to induce target cell apoptosis, while Th1 cells activate macrophages via IFN-γ for intracellular pathogen clearance.[1][3][6][10] In disease, these cells drive protective immunity against viruses, bacteria, parasites, and cancers but can contribute to pathology in autoimmunity, chronic inflammation, or transplant rejection through excessive cytokine production or autoreactivity.[1][3][5] Although not a single molecular entity like a receptor or enzyme, effector cells are leveraged in immunotherapies such as checkpoint inhibitors (e.g., targeting PD-1 on T cells) or CAR-T cells, which engineer enhanced effector functions for cancer treatment, highlighting their central role in adaptive and innate immunity.[3][5]
