Overview

Intentional allo-rejection refers to the deliberate induction or harnessing of immune responses against allogeneic (genetically non-identical) cells or tissues, with the goal of stimulating the immune system. This approach leverages the robust immunostimulatory effects that occur during allorecognition—the process by which immune cells recognize and respond to foreign major histocompatibility complex (MHC) molecules. While typically studied in transplantation as a cause of graft rejection, intentional modulation of this pathway can be used experimentally or therapeutically to boost immune activity by providing strong co-stimulation for T cell activation, inducing local inflammation, and promoting maturation/activation of APCs.
