Overview

Type I interferons (IFNs) are a family of cytokines that play a central role in the regulation of innate and adaptive immune responses. They exert their effects by binding to the type I interferon receptor (IFNAR), which is present on nearly all nucleated cells. The IFNAR is a heterodimeric cell surface receptor composed of two transmembrane subunits: IFNAR1 and IFNAR2. Upon ligand binding, JAK kinases are activated, leading to STAT phosphorylation and translocation to the nucleus to induce the expression of interferon-stimulated genes (ISGs). This pathway orchestrates broad immune responses—including potent antiviral defenses—making it a target for therapeutic intervention in infections, cancer, and autoimmune/inflammatory diseases.
