Overview

The type I interferon (IFN) receptor signaling pathway is a critical mechanism by which the immune system detects and responds to viral infections and other immune challenges. This pathway is initiated when type I interferons bind to their common cell surface receptor complex known as the type I interferon receptor (IFNAR), which consists of two subunits: IFNAR1 and IFNAR2. Upon binding of a type I interferon ligand, intracellular kinases JAK1 and TYK2 are activated, leading to phosphorylation of STATs and subsequent transcriptional activation of hundreds of interferon-stimulated genes (ISGs) that mediate antiviral defense, immunomodulation, apoptosis regulation, and anti-proliferative effects.
