Overview

"Immune system reset" denotes a therapeutic strategy and biological process whereby the existing dysfunctional or autoreactive immune repertoire is ablated or depleted—commonly by cytotoxic agents and/or immunosuppressive conditioning—followed by regeneration of naive immune populations, either naturally or via stem cell transplantation. This reset aims to induce long-term tolerance, eradicate autoreactive memory, diversify antigen receptor repertoires (TCR/BCR), and establish a stable, self-tolerant immune state. It is used experimentally and clinically in severe autoimmune diseases such as multiple sclerosis and rheumatic diseases, and is mechanistically underpinned by immune cell depletion, thymic reactivation, generation of new regulatory populations, and restoration of homeostatic immune balance. Not regarded as a molecular target, but rather as a process involving multiple cellular and molecular subsystems[3][6][4][8][1].
