Overview

Immunologic adjuvants are substances used in combination with vaccine antigens to enhance, accelerate, or prolong antigen-specific immune responses. Adjuvants exert effects through several mechanisms, such as promoting antigen uptake and presentation by APCs, inducing the secretion of cytokines, recruiting immune cells, and modulating T and B cell responses to strengthen both innate and adaptive immunity. Common adjuvants include aluminum salts, oil emulsions, and PAMP-based agents. These are critical components in modern vaccines (for infectious diseases and cancer immunotherapy), compensating for weak immunogenicity of purified antigens and ensuring effective, long-lasting protection. While effective, they must be carefully formulated to balance efficacy with safety due to possible inflammatory or allergic reactions.
