Overview

Immune-targeting is a broad therapeutic concept rather than a specific molecular target like a receptor or enzyme (National Cancer Institute, 2023). It refers to the strategic use of agents to modulate the immune system's activity to treat various conditions, most notably cancer and autoimmune diseases (Nature Reviews Drug Discovery, 2020). In oncology, immune-targeting therapies such as checkpoint inhibitors aim to restore the ability of T-cells to recognize and destroy malignant cells (PubMed, PMID: 30308138). Conversely, in inflammatory and autoimmune conditions, the goal is often to suppress specific pathways or deplete specific immune cell populations to reduce tissue damage (NIH, 2022). Because "immune-targeting" describes a class of interventions rather than a single biological entity, it lacks a specific molecular classification or gene association. It represents a diverse field of pharmacology that includes monoclonal antibodies, vaccines, and cell-based therapies (StatPearls, 2023). This approach has revolutionized the treatment of advanced malignancies and chronic inflammatory disorders by focusing on the host's immune response rather than the disease cells directly (Cell, 2019).

Mechanism of action

Immune-targeting involves various mechanisms including checkpoint inhibition (blocking PD-1/PD-L1 or CTLA-4), antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and cytokine modulation to either enhance or suppress immune activity (Nature Reviews Drug Discovery, 2020; PubMed, PMID: 30308138).
