Overview

Immunoglobulin E (IgE) specific for Alternaria alternata allergens is a specialized class of antibodies that mediates allergic reactions to the common environmental mold Alternaria alternata. This fungus is widely recognized as a potent source of aeroallergens, with Alt a 1 being its primary clinical component (Source: Gabriel et al., 2016, PMID: 26833343). In sensitized individuals, these specific IgE antibodies bind to high-affinity receptors (FcεRI) on the surface of mast cells and basophils. Upon subsequent exposure to Alternaria spores, the allergens cross-link the surface-bound IgE, triggering the immediate release of inflammatory mediators such as histamine, proteases, and cytokines. This biological process is a significant driver of allergic rhinitis and is strongly associated with the development of severe, steroid-resistant asthma and acute asthma exacerbations (Source: Bush and Prochnau, 2004, PMID: 15242831). Therapeutic interventions, most notably the monoclonal antibody omalizumab, target the IgE molecule itself to reduce the levels of free circulating IgE and downregulate receptor expression, effectively mitigating the allergic response (Source: StatPearls, NBK545184).

Mechanism of action

Monoclonal antibodies bind to the Fc region of circulating IgE, preventing its binding to the high-affinity IgE receptor (FcεRI) on mast cells and basophils, thereby inhibiting the allergic inflammatory cascade (Source: PubMed, PMID: 12743344).
