Overview

Immunoglobulin E (IgE) specific for cross-reactive invertebrate tropomyosins is a primary mediator of allergic hypersensitivity across diverse species, including crustaceans, mollusks, insects, and arachnids (Reese et al., 1999, PMID: 10352397). Tropomyosin is a highly conserved muscle protein that functions as a pan-allergen, leading to significant clinical cross-reactivity, such as the mite-cockroach-shrimp syndrome (Santos et al., 2015, PMID: 26334552). These specific IgE antibodies bind to high-affinity FcεRI receptors on mast cells and basophils; upon subsequent allergen exposure, receptor cross-linking triggers the release of inflammatory mediators like histamine and leukotrienes (Lopata et al., 2010, PMID: 20549798). This physiological response results in clinical conditions such as food allergy, asthma, and potentially fatal anaphylaxis (NIH/NIAID Food Allergy Guidelines). Therapeutic approaches targeting these IgE molecules include the use of monoclonal antibodies like omalizumab, which sequester circulating IgE to prevent receptor binding, and allergen-specific immunotherapy (AIT), which aims to induce immune tolerance and the production of protective IgG4 antibodies (WAO Journal, 2015, PMID: 25905002).
